CHARACTERIZATIONThe daffodil lectin is present in all vegetative tissues. However, the lectin concentration is developmentally regulated. In resting bulbs the daffodil lectin represents about 5-10% of the total protein. Lectin activity has been detected in more than 25 species and varieties of Narcissus including the botanical species: N. biflorus, N. conspicus, N. dub. campernelle, N. jonquilla, N. lobularis, N. maclaeyi, N. moschatus cernus, N. nanus, N. obvallaris, N. odorus regulosus, N. ornatus, N. physaloidus, N. pumilus, N. recurvus and N. tazetta. BIOACTIVITYNPA gave strong precipitation reactions with several yeast mannans, but did not precipitate with alpha-D-glucans (e.g., dextrans and glycogen). The daffodil lectin agglutinates rabbit but not human erythrocytes. See: Phys Plantarum 1988 (73);52-57 The lectin is virtually non-mitogenic towards human mononuclear cells. See: Lectin Biol Biochem Clin Biochem 1990 (7); 259-263 The daffodil lectin inhibits infection of MT-4 cells by HIV-1, HIV-2 and simian immunodeficiency virus (50% effective concentration being 0.5-0.6 gg/ml) at concentrations comparable to those of dextran sulphate inhibition of these viruses. NPA did not inhibit the replication of other enveloped viruses except for human cytornegalovirus. NPA suppresses syncytium formation between persistently HIV-1 or HIV-2 infected HUT-78 cells and uninfected MOLT-4 cells. See: Antimicrob Agents Chemotherap 1991 (35); 410-416 The daffodil lectin was also shown to prevent rabies virus attachment to susceptible cells, and affect rubella virus multiplication after the attachment step. The daffodil lectin shows antimetabolic effects towards nymphal stages of the rice brown planthopper (Nilaparvata lugens) and the peach potato aphid (Myzus persicae) when the lectin is added to artificial diets.
